Abstract
The goal of this project is to recreate in vitro, as closely as possible, the features of the melanoma tumor in the proper microenvironmental context and to use these 3D in vitro models of metastatic melanoma to characterize the dynamics of phenotypic plasticity of primary tumors, biopsies or cell lines derived from cancer patients thus facilitating risk stratification of cancer patients and developing personalized treatment strategies. I will generate 3D in vitro models of metastatic melanoma that are capable of 'mapping' extracellular gradients of lactate and O2 with submicron spatial resolution, to establish whether direct quantitative readouts of these gradients relate to biologically-relevant changes in cell response to individual therapies, going beyond the possibilities offered by current standard chemosensitivity assays.
