Abstract

Brain ischemia is the second cause of death and the leading cause of disability worldwide. Despite this, tissue plasminogen activator is the only pharmacological treatment available, and its use is limited to <10% of patients, being effective only within 3-6 hours from symptoms onset. These alarming data require that every intervention be implemented early. Among promising markers for an early diagnosis, a relevant role is played by microRNAs (miRNAs), small RNA molecules of 20-22 nucleotides capable of binding numerous messenger RNAs, promoting their degradation.
The aims of this project will be: (1) the study of the expression profiles of a miRNA group, involved in the pathogenesis of the ischemic damage and released into the circulation after cerebral ischemia; (2) the identification of the specific miRNAs released by neurons after stroke induction and their role in inducing microglia/macrophages polarization; (3) the development of a medical device for a direct and fast miRNA detection and of a chip for cross-talk investigation between engineered tissues.
Combined monitoring of different miRNAs will shorten the timing of disease prognosis and diagnosis.
