Abstract
We offer N-terminal protein/peptide sequencing using a Shimadzu PPSQ 33B sequencer at IBBR-Napoli. The method is based on the Edman degradation reaction, which labels the N-terminal amino acid residues within a polypeptide such that they can be cleaved from the chain in a stepwise manner. The benefits of using Edman degradation for protein sequencing are:
o Guaranteed N-terminal sequence of proteins;
o No sample pre-treatment required:
o Differentiation of isobaric amino acids (leucine and isoleucine):
o Sequencing of unknown proteins not registered in databases.
