Abstract

The need for new strategies to improve the understanding of cancer mechanisms and translate it into personalized therapeutic strategies is more and more urgent. A viable approach to address these problems is to take advantage of pharmacogenomic information and focus attention on modeling global cancer pathways as connectivity maps that link multiple signaling pathways. Such approaches can ultimately identify hub proteins integrating multiple mechanisms of cell proliferation, survival and adaptation. Nodal proteins act by establishing simultaneous interactions with multiple ligands, aggregating molecular networks that affect the bioavailability and activity of each interactor, and determining the final specific disease phenotype. The overall goal of our project focuses on the development of a novel structural pharmacogenomic pipeline that advances the mechanistic understanding of the roles of Hsp90 networks in determining cancer phenotypes into the discovery and design of molecular tools to improve cancer drug development, with a clear impact on patient stratification, therapy and treatment efficiency.

Goals

The overall objective will be achieved through the following specific goals, designed to integrate strategies addressing different intertwined questions:
1) Establishment of a novel computational pharmacogenomic strategy to rationalize the role of HSP interaction networks in specific cancer types.
2) Characterization of fundamental aspects of Hsp90 networks/interactions for different cell compartments and cancer types.
3) Development of chemical tools to inhibit/modulate the functional properties of Hsp90 networks.
