Th1 and CD4+ cytolytic T cell differentiation in a murine model of Chagas Disease (American trypanosomiasis)

Cytotoxic CD4+ T cells (CD4CTLs) have been described both in humans and mice, but their role in host protection against infectious disease, as well as the mechanisms that lead to their differentiation, are not clearly understood yet. Maria Bellio, from the Instituto de microbiologia Paulo de Góes, Universidade Federal do Rio de Janeiro (Brazil) will give a seminar on  recent results from her group showing that CD4+ TCRαβ+ T lymphocytes expressing high levels of granzyme B (GzB) can be found in the spleen and in the myocardium of mice infected with the intracellular parasite Trypanosoma cruzi. These cells display Ag-specific cytolytic activity in vitro and also express CD244 and CD107a, a cytotoxic and degranulating marker, respectively, as well as the ThPoK, T-bet and Runx3 nuclear factors. Moreover, as prof Bellio's group recently demonstrated for the Th1 response, they found that the T-cell intrinsic IL-18R/MyD88 signaling is a crucial element also for the induction of CD4CTLs in this infection model.
