Abstract

In this project we will investigate mechanisms leading to autoimmune diabetes (AD). Findings will be explored and validated in another autoimmune disease related to AD, rheumatoid arthritis. By taking advantage of an available large biobank and liquid biopsies from AD patients of different ages and various degree of bcell loss, we pursue the following specific aims: 1)To study the role of gene polymorphisms and autoantibodies associated with AD progression or protection; 2)To study cell-mediated/innate immunity and neutrophils; 3)To evaluate the role of miRNAs as regulators and biomarkers of bcell identity, islet function and dysfunction; 4)To identify metabolomics markers of gluco-lipotoxicity and bcell damage; 5)To evaluate the effect of gluco-lipotoxicity and pro-inflammatory cytokines on human bcells for identifying biomarkers of bcell damage and recovery; 6)To investigate the role of oxidative posttranslational modifications (oxPTM) of bcell autoantigens in AD pathogenesis, diagnosis and prognosis.
