Abstract

The keystone of Celiac Disease (CD) pathogenesis is an altered immune response to gluten in genetically predisposed individuals. At present, the only therapy for CD patients is a permanent gluten-free diet (GFD). Compelling evidences support the central role of IL-10 and IL-10-secreting regulatory cells, including tolerogenic dendritic cells (DC) and T cells, in gut homeostasis and in controlling inflammatory responses to dietary antigens (Ags). We propose to determine the presence and functions of IL-10 and IL-10-related regulatory cells in CD patients at different stage of the disease (potential CD, CD patients at onset, compliant to GFD), and to generate stable and effective tolerogenic DC suitable for cell-therapy in CD by means of Lentiviral Vectors co-encoding for gluten-Ags and tolerogenic molecules (IL-10 or IDO). Results will provide new insights into the immunological mechanisms underlying CD progression and response to GFD, and will open new perspectives for the cure of CD.
