Multimodal optical microscopy for tissue characterization and diagnostics

Riccardo Cicchi (from the National Institute of Optics [Cnr-Ino], Sesto Fiorentino, Italy) will give a Seminar on 'Multimodal optical microscopy for tissue characterization and diagnostics'.

Modern laser scanning micro-spectroscopy techniques offer promising solutions for tissue imaging at the subcellular level as they can provide morphological, functional, chemical and mechanical information without the use of exogenous labels. In particular, two-photon fluorescence (TPF) microscopy can be used for selectively exciting fluorescence from endogenous fluorophores, allowing for a detailed label-free morphological characterization of biological tissues. The possibility to specifically excite fluorescence from NADH and FAD nucleotides extends the capability to morpho-functional characterization through the non-invasive assessment of tissue metabolic condition. Second-harmonic generation (SHG) microscopy is a coherent microscopic modality able to provide background-free images of anisotropic hyperpolarizable repetitively patterned biological molecules, such as collagen, muscle, or microtubules. SHG microscopy can be used for the investigation of structural organization and fibrillar orientation of collagen-rich tissues such as human dermis, keloid, fibrosis, thermally-treated samples, and tumour microenvironment. Vibrational microscopy techniques, such as spontaneous Raman and Coherent-Antistokes Raman Scattering (CARS) microscopies, provides label-free chemical contrast of a biological specimen thanks to their sensitivity to molecular vibrations, allowing for a morpho-chemical characterization. Brillouin micro-spectroscopy, exploiting photon-phonon scattering, provides a powerful label-free microscopic modality for the contact-less non-invasive assessment of tissue visco-elastic properties, useful for a morpho-mechanical characterization of the specimen. The combination of multiple of the above-mentioned imaging techniques in the same microscope allows integratingmorphological with functional and mechanical information, providing a high-resolution label-free alternative to both histological and immunohistochemical examination of tissues. Even if a lot of work needs to be done before these methodologies are recognized as a diagnostic standard, these techniques offer the potential to find a stable place in a medical setting in the near future.
