Functional genomic screens of sister chromatid cohesion and cell cycle control

Rob Wolthuis’ group (VUmc Medical Faculty Department of Clinical Genetics Section of Oncogenetics Amsterdam, The Netherlands) studies the regulation of the cell cycle in relation to genetic disorders. An important object of study are the so-called cohesinopathies, diseases related to defects in the regulation of sister chromatid cohesion. Such defects may relate to mutations or loss of function of genes that regulate S-phase, mitosis, or the actual components of cohesin rings that entrap replicated sister chromatids. Functional genomic screens are used to identify factors that connect sister chromatid cohesion to the cell cycle and to identify potential drug targets for treatment of cancer cells that display cohesion defects.
