Abstract

The project aims at establishing an innovative platform to study intrinsic mechanisms and extrinsic signals that define the cross-talk between prostate cancer cells and infiltrating fibroblasts during PCa progression. The platform includes stromal cells culturing from radical prostatectomies (normal and cancer associated fibroblasts), development of 3D organoids from primary PCa cells and 2D non-transformed cell lines, both genetically engineered to recapitulate PCa tumorigenesis. These models will be used to study oncogenic mechanisms due to cell-intrinsic (genetic and epigenetic) and extrinsic (tumor microenvironment) processes, and their interaction during PCa onset and progression.
Validation of key molecular determinants will uncover potential therapeutic targets, and allow the design of lab-on-chip kits as diagnostic tools

Goals

Task 7.1. Real time monitoring of proliferation, motility, invasion.
Task 7.2. On-chip detection of selected proteins from onco-secretome.
Task 7.3. Realization of a prototype for clinical applications.
