Shedding light on nuclear pore assembly during Drosophila embryogenesis

Dr. Paolo Ronchi (Electron Microscopy Core Facility, EMBL, Heidelberg, Germany) will give a Seminar on 'Shedding light on nuclear pore assembly during Drosophila embryogenesis'.

Nuclear pore complexes (NPCs) span the nuclear envelope (NE) and mediate nucleocytoplasmic transport. In metazoan oocytes and early embryos, NPCs have been observed not only within the NE, but also at some endoplasmic reticulum (ER) membrane heets, termed annulate lamellae (AL).
In collaboration with the lab of Martin Beck at EMBL, Paolo Ronchi's group demonstrated by correlative light and electron microscopy that annulate lamellae provide the source of NPCs necessary for the expansion of the nuclear envelope during the rapid growth of the nuclei in the early development of drosophila embryos. Further experiments showed that the assembly of pores into ALs occurs from precursor granules that organize in the oocyte as phase separated organelles and interact to give rise to the final structure.
By FIBSEM, they could characterize the 3D organization of these structures, revealing interesting common morphological features.
