- Abstract
- In animal models, we will create enduring functional alteration within DAT and/or SERT function and will be will be scored for ability to reproduce ADHD symptoms, according to the following endpoints. Behavior: circadian activity; impulsivity; attention; anxiety, motivation spatial memory. Neurochemistry: At sacrifice, the same areas will be assessed (gene expression and protein density) for DAT, SERT and selected subtypes of DA and 5-HT receptors.
Therapeutic endpoints - We will evaluate the possibility to exploit the serotonin (5-hydroxytryptamine, 5-HT) receptor 7 (Htr7) as a target for a psycho-pharmacological approach. This choice is based on recent preliminary evidence of a functional relationship between Htr7 and rats' impulsive behaviour.
Diagnostic endpoints We will measure in plasma samples from ADHD children the levels of aAbs against DAT and/or SERT, using specially tailored peptide fragments as antigens in the ELISA. RTP analysis will be used to evaluate markers variation in blood samples in normal and ADHD children Neural components are a validated biomarker in several neuro-behavioural syndromes. (literal)
