Little information is available on the fundamental nature of stemness, the stability of cell differentiation, and cell reprogramming from in vivo systems. Novel insight may be useful to the study of senescence and the molecular controls of cell differentiation in non-mammalian, underinvestigated models. The hydrozoan "immortal" medusa Turritopsis dohrnii challenges basic aspects of both aging theories and cell reprogramming: life cycle reversal can occur at any stage of medusa growth, including the adult. Reverse development (RD) is actually induced by epigenetic factors as well as by endogenous signals of senescence. This unparalled potential in the animal kingdom is achieved by cell dedifferentiation-redifferentiation and transdifferentiation mechanisms. Differently, the colonial tunicate Botryllus schlosseri weekly exhibits a cyclical, zooid budding rhythm and constitutive generation of the whole colonial body (blastogenesis, BG), with regular reactivation of early development molecular programs, differentiation of pluripotent stem cells, and putative transdifferentiation of adult somatic cells. These extraordinary potentials for regeneration and dedifferentiation represent ideal models to understand how gene regulatory networks and their associated cell behaviors drive ontogeny, the molecular switches of developmental transitions, and the mechanisms controlling the stability of cell fate. PHENIX aims to shed light on the molecular pathways involved in RD of T. dohrnii and BG of B. schlosseri, focusing on the mechanisms controlling the ontogenetic direction of life cycle, the stability of cell fate, and stem cell renewal. The workplan will embrace transcriptomic, gene expression and morpho-functional analyses. The comparison of these processes in two evolutionary distant invertebrate models, a cnidarian and a chordate, will ensure the identification of the evolutionarily most conserved genes/pathways underlying theses processes. Transcriptome profiling will be used: (1) in each model species, to identify genes differentially expressed among specific life cycle stages characterized by the occurrence of the above mentioned processes; (2) to identify genes and pathways up- and down-regulated in these processes in both our models. Among the identified differentially expressed genes, we will focus our attention on: the four Yamanaka's factors (Sox2, Klf4, Oct3/4, cMyc), transcription factors with top reprogramming activity in vertebrates; YAP/TAZ, the regulators of the Hippo signaling pathway; the gap junctions (GJs) machinery. Pharmacological perturbation of cell-cell communications, regeneration-dedifferentiation experiments, morpho-functional analyses and gene expression studies will contribute towards a better understanding of the extraordinary biological phenomena of RD and BG.

The objective of the GoJelly project is to develop, test and promote a gelatinous solution to microplastic pollution by developing a TRL 5-6 prototype microplastics filter (GoJelly) for commercial and public use, where the main raw material is jellyfish mucus. In doing so, the consortium addresses two environmental issues with one approach by removing the commercially and ecologically destructive sea and coastal pollution of both jellyfish and microplastics. This innovative approach will ultimately lead to less plastic in the ocean, municipal demand (and thereby competitive prices) for jellyfish raw material to fill the "mucus-need" by filter developers, and in turn more jobs for commercial fishers in offseasons. The by-products of the GoJelly biomass have other uses as well, ensuring that GoJelly also delivers a green innovation, resulting in novel, valuable resource for the food and feed industry as well as agro-biological fertilizer for organic farming. The GoJelly prototype products will be tested and demonstrated in three different European seas (Norwegian, Baltic and Mediterranean), by a range of stakeholders, including commercial fishers and industry partners. Tying it together, the project will also ensure the possibilities for broader European promotion and utilization of GoJelly at the local, regional and global level by delivering a socio-ecological methodological toolbox for forming and implementing policies. GoJelly will broadly communicate its results in several formats such as traditional social media, open lab ship cruise, and in the form of an experimental online game depicting different management scenarios under different jellyfish- and microplastics combinations. An interdisciplinary and international consortium consisting of technology developers, business analysts, fishing companies, research institutes, and both natural and social scientists will realize GoJelly, and will ensure the uptake of GoJelly products by industry and policy makers.

Studies of biochemical / nutraceutical /microbiological and nutritional features of Mediterranean and European jellyfish for their exploitation as food or food ingredient. The exploitation of jellyfish biomasses is considered as an opportunity to produce new products.

Study aimed to the recovery and utilization of biophenols from Olea europea by-products, with the general objectives to promote the scientific cooperation between Italian and Greek Scientific Institutions and to improve the knowledge and the management of the olive by-products considering two main aspects: the recovery of high value compounds and the environmental impact. My activity is devoted to the evaluation of the biological activity of polyphenols from OMWW using in vitro assays and cell culture systems (WP5 action 5.3.1 Biological assay and data evaluation). Cytotoxicity tests and the effects on cell proliferation and apoptosis of cell cultures of Human Epidermal Keratinocyte (HEKa) were performed on HEKa treated with polyphenols and ultrafiltered OMWW in UV-irradiated cells versus non-irradiated cells. The ability to protect skin from UV-induced DNA damage was evaluated. The OMWW based products are targeted to cosmetic and/or nutraceutical industry, representing an ideal applicative field for the formulated products developed in project activity.

Study of the role of Gap-Junction-mediated-Intercellular-Communications (GJIC) and of retinoic-acid in the embryo-development and polyp-regeneration of the model Clava multicornis, by using on site facilities at the Station Biologique de Roscoff. The GJ and RA roles were investigated by pharmacological-inhibition using GJ-inhibitors, RA and RA-inhibitors looking at morphogenesis, differentiation and regeneration ability as well as at specific behavior, gene and protein expression in marine invertebrates. The putative presence of GJ-hemi-channels was analyzed by using GJ-permeant and a non-permeant fluorescent dyes and observation at confocal microscopy. The in vivo experiments allowed to clarify the GJIC mediated key mechanisms with high level of functional conservation from basal invertebrates to chordates and could be the bases for knowledge of the GJ roles in physiological or pathological conditions in mammalian cells.
