A wide range of\nvariation in plant and plant organ morphology was found in the TILLING collection formed by about 1900 lines with DNA\n(M2 generation) and seed (M3/M4 generation). The estimated rate of mutation, on the basis of the screening for three\ngenes of interest, was 1 mutation/Kbp/400 plants.\nTwo of these genes are involved in the biosynthetic pathway of plant secondary metabolites (triterpenic saponins and\ntrypsin inhibitors) displaying physiological, agronomical and industrial interest. In particular, two segregant mutant lines\nof the 6 found for the orthologue of the trypsin inhibitor gene MsTI from M. scutellata were examined but no significant\nvariation in trypsin inhibition was found compared to the full sib wild type. Two lines carrying different point mutations in\nthe MtPHY1 gene, coding for an extracellular phytase, were also studied, one of them showing different (P = 7.5%)\nenhanced phytase activity with respect to the control line.\nTransposon mutagenized lines were also produced (approx. 1500 R0 plants) according to the strategy of d'Erfurth et al.\n(2003). Several mutants were isolated also regarding plant architecture (for example leaf and flower morphology) and\nsecondary compounds (tannins).
