Moreover, we showed that isch-emic\nconditions induced specific neuronal loss not only in\nhippocampal, but also in cortical and striatal organotypic\ncultures and the P2 receptor antagonists basilen blue and\nsuramin prevented these detrimental effects. In the in vivo\nexperiments we confirmed the induction of P2X receptors in\nthe hippocampus of gerbils subjected to bilateral common\ncarotid occlusion. In particular, P2X2 and P2X4 proteins be-came\nsignificantly up-regulated, although to different extent\nand in different cellular phenotypes. The induction was con-fined\nto the pyramidal cell layer of the CA1 subfield and to the\ntransition zone of the CA2 subfield and it was coincident with\nthe area of neuronal damage. P2X2 was expressed in neuro-nal\ncell bodies and fibers in the CA1 pyramidal cell layer and\nin the strata oriens and radiatum. Intense P2X4 immunofluo-rescence\nwas localized to microglia cells.\nOur results indicate a direct involvement of P2X receptors\nin the mechanisms sustaining cell death evoked by metabo-lism\nimpairment and suggest the use of selected P2 antago-nists\nas effective neuroprotecting agents."@en ; prodottidellaricerca:prodottoDi istituto:CDS059 ; pubblicazioni:autoreCNR unitaDiPersonaleInterno:MATRICOLA22471 . ns10:ID338149 pubblicazioni:rivistaDi prodotto:ID4550 .
