The final injection solution contained\n230 mM sodium [1-13C]pyruvate, 100 mM TRIS\nbuffer, 0.27 mM Na2EDTA and 20 microM Dotarem\n(Guerbet, Paris, France). Temperature of solution was\nabout 37\u00BAC and pH = 7.6. A dose of 20 mL was administered\nover 10 s by manual injection.\nMR studies\nThe mini-pigs underwent both 1H MR imaging and hyperpolarized\n13C MRS. The experiments were performed with\na 3 T GE Signa HDx (GE Healthcare, Waukesha, WI, USA)\nscanner with a 13C quadrature birdcage coil (Rapid Biomedical,\nW\u00FCrzburg, Germany). Anatomical imaging was\nacquired with the body coil and FIESTA sequence\n(FOV=35, FA=45, TE/TR=1.71ms/3.849ms).\n13C dynamic spectra were acquired using elliptic-FIDCSI\npulse sequence (bandwidth 5000Hz, 2048 pts, 10\u00BA FA). A\nlong-axis slice of 20 mm was selected during excitation.\nSpectra covering the heart were acquired from the beginning\nof the injection of the hyperpolarized [1-13C]pyruvate,\nevery 2 s, for 120 s. Data processing\nData processing was performed by using MATLAB (The\nMathworks, Inc., Natick, MA) and jMRUI software tools.\nThe dynamic spectra were phase corrected by adjusting\nboth zero-and first-order frequency-dependent phase\ncomponents. Pyruvate, pyruvate hydrate, alanine, lactate,\nand bicarbonate were estimated by using AMARES algorithm\nincluded into jMRUI tools. Accordingly, time\ncourses of metabolites are generated.\nResults and discussion\nSpectra obtained from the dynamic acquisition are shown\nin Figure 1. The [1-13C]pyruvate, [1-13C]pyruvate hydrate\nand metabolites peaks ([1-13C]lactate, [1-13C]alanine, and\n13C-bicarbonate) have been detected and plotted.
