The occurrence of DNA strand\nbreakage damage was evaluated all year round using the comet assay.\nSemen of buffaloes A and B was collected each month from December\nto November. Ejaculates were assessed using a computer sperm\nanalyzer. Spermatozoa (106/ml) embedded in low melting point\nagarose on a microscope slide were lysed, DNA was unwound and\nelectrophoresis carried out (pH 11). Four replicates were performed\nfor each sample. DNA was visualised after staining with ethidium\nbromide, 200 sperm cells per slide were scored for the presence and\nabsence of comets (DNA-damaged cells). The percentage differences\namong months and subjects were assessed by ANOVA. Comet percentages\nwere higher in June (A = 40 \u00B1 2%; B = 20 \u00B1 4%) and July\n(A = 9 \u00B1 1%; B = 44 \u00B1 2%) than in April (A = 5 \u00B1 1%;\nB = 8 \u00B1 1%) and September (A = 2 \u00B1 1%; B = 1 \u00B1 2%).\nObserved values were significantly different (p < 0.01) in June (buffalo\nA) and July (buffalo B) compared with the overall trend. Our data\nsuggest an increase of DNA damage in the summer period (despite an\nobserved increase in blood testosterone values), with a slight time shift\nbetween subjects.
